There's a PD program called GPaint which is really good, and has lots of features. It runs on a 464 too. No idea about a mouse though. It's not in English but the icons represent their functions well, and you can always just use trial and error, if you don't understand German.

On the CPC back in the day I used Advanced OCP Art Studio.

If you want to create pixel art on a PC/Mac, for free you could use GrafX2 (although that went too far with the retro styling).

For animated sprites Asesprite is popular, and not expensive.

You probably can't go wrong with WinUAE and DPaint/PPaint either.

If you're the proud owner of an x-Mem, you could also use iMPdraw (http://www.cpcwiki.eu/forum/programming/impdraw-v1-0f/) which will allow you to make fullscreen graphics in mode 0!

Personally I would suggest to use IMPdraw, that's modern and has all features I need. The other one which I use is the GPaint (as suggested from Egotrip, see before), because it does support the Atari ST Mouse (which I can use from my Atari ST computer. For the CPC it need a little adapter, see Wiki).
